Three days ago, I first got this error after about 2 minutes of play.
What could this be?

That would be a unique error report related to the crash of your game. If you submit that following the pop-up’s instructions, we can have a look and see what might have happened.
To start with though, please go through the steps listed in the Diablo III Locked Up support article. Please follow the steps listed thoroughly and see if that improves things for you.

My problem was resolved by deleting “D3Prefs.txt” file.
I played for about an hour without any problems.
