Loot Specification

I am not sure how this works so I ask here.

I go into a dungeon, kill a boss, select my loot specification, eg healer, tank, dps and then I loot.

Will that work? Or does it lock me to the loot spec I was before I entered the dungeon? Or maybe the loot spec I was when I engaged the boss?

You have to choose your loot spec Before you engage the boss.
You can change your loot spec on a boss by boss basis.
